Born in New York City to a Juilliard-trained pianist father and a vocalist mother, Frusciante's early life was immersed in music. Following Slovak's tragic death in 1988, an 18-year-old Frusciante successfully auditioned to join the band. He made his recorded debut on Mother's Milk (1989), but it was the band's subsequent collaboration with producer Rick Rubin on Blood Sugar Sex Magik (1991) that catapulted them to international stardom. Overwhelmed by the sudden fame, Frusciante quit the band in 1992.

In a surprise to fans, Frusciante rejoined the Red Hot Chili Peppers in 2019, marking his third stint with the band.
